چکیده

MRI contrast properties of self-aggregated nanoparticles made from GdIII and 1,2,4,5-tetrakis(pyrazol-1-yl-methyl-3-carboxylate)benzene (L) are investigated. Viscosity of GdL2 suspensions and size-characteristics of GdL2 nanoparticles allow an estimate of their large rotational correlation time. Moreover, two to three H2O molecules are bound, on average, to GdIII ions, as deduced from 17O NMR titration of the DyIII analogue. The large relaxivity of the particles, along with the prominent peak in the range 20–60 MHz, are the consequence of these two properties. Longitudinal (r1) and transverse (r2) relaxivities are determined as a function of monomer concentration at 20 ◦C and 20 MHz. The ratio r1/r2 appears to be favorable for MR imaging using T1-weighted gradient echo sequences. According to preliminary tests conducted under physiological conditions, the GdL2 nanoparticles have some potential as contrast agent provided their stability can be increased. © 2003 Elsevier B.V. Contrast agents for magnetic resonance imaging synthesized with ring-opening metathesis polymerization.

A monomer for ring-opening metathesis polymerization (ROMP) has been developed that also functions as a portion of a GdIII chelating moiety for a magnetic resonance imaging contrast agent. An increase in per GdIII relaxivity was shown upon transition from monomer to polymer. Additionally, extremely large molecular relaxivities were achieved through incorporation of multiple GdIII ions per polym...

Surface PEG Grafting Density Determines Magnetic Relaxation Properties of Gd-Loaded Porous Nanoparticles for MR Imaging Applications

Surface PEGylation of nanoparticles designed for biomedical applications is a common and straightforward way to stabilize the materials for in vivo administration and to increase their circulation time. This strategy becomes less trivial when MRI active porous nanomaterials are concerned as their function relies on water/proton-exchange between the pores and bulk water. Here we present a compre...

Effect of coating thickness of iron oxide nanoparticles on their relaxivity in the MRI

Objective(s):Iron oxide nanoparticles have found prevalent applications in various fields including drug delivery, cell separation and as contrast agents. Super paramagnetic iron oxide (SPIO) nanoparticles allow researchers and clinicians to enhance the tissue contrast of an area of interest by increasing the relaxation rate of water. In this study, we evaluate the dependency of hydrodynamic si...

ANALYTICAL STUDY OF EFFECT OF BILAYER INORGANIC AND ORGANIC COATING AROUND THE IRON OXIDE NANOPARTICLES ON MAGNETIC RESONANCE IMAGING CONTRAST

Background & Aims: In recent years, iron oxide nanoparticles have been used in contrast-enhanced magnetic resonance imaging for diagnosing a wide range of diseases. In order to provide biocompatibility and prevent the toxicity of the nanoparticles, using organic or inorganic coating around these nanoparticles is important for their application. The aim of this study is to investigate the effect...

Optimizing the relaxivity of GdIII complexes appended to InP/ZnS quantum dots by linker tuning.

Three bimodal MRI/optical nanosized contrast agents with high per-nanoparticle relaxivity (up to 2523 mM(-1) s(-1) at 35 MHz and 932 mM(-1) s(-1) at 200 MHz) have been prepared connecting up to 115 tris-aqua Gd(III) complexes to fluorescent non-toxic InP/ZnS quantum dots. The structure of the linker has an important effect on the relaxivity of the final multimeric contrast agent.
